چکیده

Although synaptotagmin I, which is a calcium (Ca 2+)-binding synaptic vesicle protein, may trigger soluble N-ethylmaleimide-sensitive factor attachment protein receptor (SNARE)-mediated synaptic vesicle exocytosis, the mechanisms underlying the interaction between these proteins remains controversial, especially with respect to the identity of the protein(s) in the SNARE complex that bind(s) to synaptotagmin and whether Ca 2+ is required for their highly effective binding. To address these questions, native proteins were solubilized, 2+-independent 3 fashion, providing biochemical evidence that synaptotagmin I binds SNARE complexes before Ca 2+ influx into presynaptic nerve terminals.

Synaptotagmins I and IV promote transmitter release independently of Ca binding in the C2A domain

At nerve terminals, a focal and transient increase in intracellular Ca triggers the fusion of neurotransmitter-filled vesicles with the plasma membrane. The most extensively studied candidate for the Ca-sensing trigger is synaptotagmin I, whose Cadependent interactions with acidic phospholipids and syntaxin have largely been ascribed to its C2A domain , although the C2B domain also binds Ca (re...

Localization of synaptotagmin-binding domains on syntaxin.

Synaptotagmin, an abundant calcium- and phospholipid-binding protein of synaptic vesicles, has been proposed to regulate neurotransmitter release at the nerve terminal. To understand better the biochemical mechanism of neurotransmitter release, we have investigated the calcium-dependent and -independent protein-protein interactions between synaptotagmin I and syntaxin 1a, a subunit of the recep...

The AP2 binding site of synaptotagmin 1 is not an internalization signal but a regulator of endocytosis

One characteristic linking members of the synaptotagmin family to endocytosis is their ability to bind the heterotetrameric AP2 complex via their C2B domain. By using CD4/synaptotagmin 1 chimeras, we found that the internalization signal of synaptotagmin 1 lies at the extreme COOH-terminus of the protein and can function in the absence of the C2B domain that contains the AP2 binding site. Howev...

Ca2+ Regulates the Drosophila Stoned-A and Stoned-B Proteins Interaction with the C2B Domain of Synaptotagmin-1

The dicistronic Drosophila stoned gene is involved in exocytosis and/or endocytosis of synaptic vesicles. Mutations in either stonedA or stonedB cause a severe disruption of neurotransmission in fruit flies. Previous studies have shown that the coiled-coil domain of the Stoned-A and the µ-homology domain of the Stoned-B protein can interact with the C2B domain of Synaptotagmin-1. However, very ...

-independent SNARE Complex Interaction

Synaptotagmins are synaptic vesicle-associated, phospholipid-binding proteins most commonly associated with Ca 1 2 -dependent exocytotic and Ca 1 2 independent endocytotic events. Synaptotagmin III is a 63.2-kD member of the synaptotagmin homology group; one of its characteristic properties is the ability to bind divalent cations and accessory proteins promiscuously. In the cytosolic portion of...
